Description :
Platelet-activating factor (PAF) is an important lipid mediator involved in inflammation. Platelet-activating factor acetylhydrolase (PAF-AH) is an extracellular phospholipase A2 which hydrolyzes the acetyl group at the sn-2 position of phospholipids.PAF deacetylation by PAF-AH causes loss of biological activity, making PAF-AH potential useful as an anti-inflammation therapy. Human recombinant PAF-AH was characterized using 2-Thio PAF as a substrate, which upon hydrolysis by PAF-AH releases a free thiol that reacts with 5,5-dithio-bis(2-dinitrobenzoic acid) (DTNB) to produce 5-thio-2-nitrobenzoic acid (TNB) with an absorbance maximum at 412 nm.
